Exercise 2 – Investigating Photosynthesis

In this exercise, you will design a study to investigate factors that may influence rates of photosynthesis. Such factors include light, temperature, and CO2 concentration. Depending on your study design, you may be able reuse some parts of the setup from Exercise 1 or you may need to create a new setup by cleaning and drying the equipment.

Procedure

NOTE: For this experiment, you may assume that the more quickly that leaf disks float, the greater the rate of photosynthesis. In actuality, density of the leaf disk, venation, and concentration of the solution may play a role in disk buoyancy, not just photosynthetic rate.

1. Identify a testable variable that may influence the rate of photosynthesis. You may identify a unique variable or select a variable from the following examples:

Light intensity
Light wavelength: bulb color
Light type: natural vs. artificial
Temperature
Concentration of CO2 (concentration of the sodium bicarbonate solutions)
Plant species
2. Create a broad research question about the variable. Record the question in Data Table 3.

NOTE: An example of a research question is, “Do basil and spinach leaves photosynthesize at the same rate?” (Do not use this exact example, as your instructor may not give you credit for an original research question.)

3. Record a hypothesis in Data Table 3. Be as specific as possible; the hypothesis should directly reflect your study design.

Note: The hypothesis is an educated guess; it does not have to be correct. An example hypothesis is, “If basil leaves have a greater rate of photosynthesis than spinach leaves, then basil leaf disks will float to the top of a sodium bicarbonate solution more quickly than spinach leaf disks.”

4. Describe your experimental design in Data Table 3. Explain the equipment and materials you will need and how the experiment will be set up.

5. Set up the experiment. Reuse materials from Exercise 1 as allowable; use clean, dry equipment where necessary.

6. Report your data in the answer panel titled Data and Results. Include any graphs in panel titled Graphs: Experimental Results.
