In an initial experiment an enzyme-catalyzed reaction was carried out with the substrate concentration a thousand times greater than the Km for that substrate. After 9 minutes, 1% of the substrate had been converted to product, and the amount of product formed in the reaction mixture was 12mmol.
In a second experiment twice as much substrate and one-third as much enzyme were combined:
(i) Calculate the rate of the reaction (mmol min-1) in the initial reaction.
(ii) Predict the effect of doubling the substrate in the second experiment and explain your prediction with reference to Michaelis-Menten kinetics.
(iii) Calculate how long it will take for 12mmol of product to be formed in the second reaction.
